چکیده

Hemin is an activator of heme oxygenase-1 (HO-1), enzyme catalyzing degradation. Up-regulation HO-1 observed in response to various pathological conditions. Moreover, pharmacological activation associated with numerous beneficial effects the organism. was shown exert, among other, anti-diabetic and anti-obesity properties. These are strongly linked adipose tissue. However, direct influence hemin on metabolism fat cells have not been explored. The present study aimed determine short-term primary rat adipocytes. We focused processes directly related lipid accumulation, such as lipogenesis lipolysis. For this purpose, isolated were subjected for 2 h 40 µM hemin, compound insulin-stimulated glucose conversion lipids, lactate release, lipolysis induced by stimuli, also antilipolytic action insulin determined. It that did affect insulin-induced release. significantly decreased stimulated epinephrine. inhibitory effect epinephrine-induced abolished presence SnMP, inhibitor HO-1, which suggests irrespective enzyme. Similar 3 12 mM glucose. reduce when replaced alanine or succinate. Apart from changes epinephrine action, it found lipolytic adipocytes isoproterenol diminished hemin. failed dibutyryl-cAMP (a protein kinase A), forskolin (an adenylate cyclase), DPCPX adenosine A 1 receptor antagonist). Additionally, be insulin, deepened results indicate exposure does metabolism, decrease adrenergic stimulation, reduced release our capable diminishing exaggerated lipolysis, occurs supraphysiological concentrations

Adrenergic receptor stimulation attenuates insulin-stimulated glucose uptake in 3T3-L1 adipocytes by inhibiting GLUT4 translocation.

Activation of the sympathetic nervous system inhibits insulin-stimulated glucose uptake. However, the underlying mechanisms are incompletely understood. Therefore, we studied the effects of catecholamines on insulin-stimulated glucose uptake and insulin-stimulated translocation of GLUT4 to the plasma membrane in 3T3-L1 adipocytes. We found that epinephrine (1 microM) nearly halved insulin-stimu...

Differences in the response of UCP1 mRNA to hormonal stimulation between rat and mouse primary cultures of brown adipocytes.

UNLABELLED Uncoupling protein 1 (UCP-1), the specific marker of brown adipose tissue, is transcriptionally activated in response to adrenergic stimuli and thyroid hormones are necessary for its full expression. We describe differences in the regulation of UCP-1 mRNA expression between rat and mouse brown adipocytes in culture, using norepinephrine (NE), triiodothyronine (T3), insulin and retino...

Lymphocyte aggregation in response to adrenergic stimulation.

A nonanemic chronic lymphocytic leukemia patient with nearly 500,000 lymphocytes/microL underwent leukapheresis when she presented with CNS symptoms and retinal vascular engorgement. Respiratory distress developed during the cell separator run, which led us to ask whether the procedure could have changed the adhesive properties of her cells. C5a desarginine, N-f-Met-Leu-Phe, adenosine diphospha...

Triiodothyronine amplifies the adrenergic stimulation of uncoupling protein expression in rat brown adipocytes.

Uncoupling protein (UCP), the mitochondrial protein specific to brown adipose tissue, is activated transcriptionally in response to cold and adrenergic agents. We studied the role of triiodothyronine (T(3)) on the adrenergic stimulation of UCP mRNA expression by use of primary cultures of rat brown adipocytes. Basal UCP mRNA levels are undetectable. Norepinephrine (NE) increases UCP mRNA during...
